Common nasal spray ingredient emerges in Massachusetts lab work on aging pathways

A common nasal spray ingredient has turned up in an unexpected corner of aging research. Early lab work suggests oxymetazoline — best known as the active ingredient in Afrin — may influence one biological process associated with aging.

Here's what to know

According to the New York Post, Dave Asprey, founder of the biohacking movement, pointed to a Northeastern University project that evaluated 6,442 drugs against 11 hallmarks of aging.

One of the compounds that drew attention was oxymetazoline, which appeared to influence intercellular communication — the signaling system cells use to send and receive messages throughout the body.

Aging can disrupt those signals, contributing to issues such as weaker immune responses and muscle and bone loss. Oxymetazoline is also found in some Sinex nasal sprays, as well as certain eye drops and creams used for redness, the outlet reported.

Still, a promising lab result is a long way from a real anti-aging treatment.

No human studies have been completed, the New York Post reported, and researchers said cell-level work on how oxymetazoline acts is underway at Harvard. Lead author Bnaya Gross cautioned, "I think we are not there yet."

Afrin is not meant for extended use.

Used briefly, oxymetazoline can relieve severe congestion, but the label says not to use it for more than three consecutive days. That limit is tied to rebound congestion, a well-known phenomenon in which repeated use can cause nasal swelling to return even worse.

Scientists studying aging are increasingly exploring whether existing drugs can be repurposed to target specific biological pathways involved in decline.

Researchers already know a great deal about how older medicines behave. Even so, many compounds that look promising in cells or computer models never prove useful, safe, or meaningful in people.

The same screening also flagged aspirin as affecting five hallmarks of aging, including intercellular communication, proteostasis, and cell senescence.

Aspirin carries known risks, including bleeding, and any off-label use should be discussed with a medical professional, the New York Post reported.

What can be done?

Afrin's intended role is short-term relief of severe nasal congestion, to be used exactly as directed.

Ongoing stuffiness may warrant evaluation for underlying allergies, irritants, or other evidence-based treatments designed for extended management.

Regular exercise, good sleep, preventive care, blood pressure control, and avoiding smoking all have strong evidence behind them, while repurposed-drug ideas such as this one still require years of testing and validation.

Screening thousands of existing drugs could help scientists identify overlooked compounds faster and at lower cost than developing entirely new ones from scratch.

As Asprey put it, "Some of the biggest longevity wins will come from a drug that's already sitting on a shelf, not from some new molecule."
